Can I replace my defective HDD 2TB, WD2002FAEX in the NAS without any problems with the WD20EFAX 2TB although it has a slower 5400 rpm but 256MB cache compared to the WD2002FAEX with 7200 rpm and 64Mb cache? Thanks

Is it only one disk in the NAS? Then you can replace it without any problem. If there are 2 disks in it, too, although 2 identical disks would of course be a better idea.
